Mrs. Mac is complaining about the PC running too slow. What is a good tool to scan it to speed it up? Or what is a good way to speed it up?

For starters:

Use MalwareBytes to scan it (www.malwarebytes.org) for malware, spyware, etc.

Use NOD32 to scan it (www.eset.com) for viruses.

Highly recommend buying Diskeeper 2011 (http://www.diskeeper.com/products/home-use/)

Always, add more RAM if possible; prices these days are very reasonable.

Close your eyes and pretend it doesn't cost $400 a 6-pack in Australia. Then pretend you are pouring yourself a tall glass and drinking. This is one beer that even the Canadians and English guys can get behind.

You could also try a good old-fashioned de-fragmentation run on the hard-drives. Oh and have a look at GameBooster for disabling non-essential processes.

Dapper, any other suggestions? I'm sure he's asking for help because he's
already tried this, but it didn't make his computer any faster. :D

I find that if I keep my cpu fan dust free I tend to run silky smooth all the time. Maybe it is in my head though. I was having problems with some of my games etc stuttering, and Chewbacca mentioned blowing out the cpu fan. It worked.

Mac, if she is having slow downs just surfing the web, maybe she's using FireFox and has a million add ons. Each add on you install on FireFox slows it down. Some more than others. 5nipers suggestions should probably be tried first.

As recommended above try CCCleaner and by the same guys MYDefrag. Clearing space off will help. Make sure VRam/swap file is set up correctly 1.5 and 2 times the amount of ram in the pc. CCCleaner will let you check your startup config so turn off all the updates and stuff that is not always used as that will slowdown start up. If TS windows XP turn off fast boot option it's crap and still takes just as long.

Also consider rather than shutting down the oc each time just use sleep or hybernate mode.

Get the free versions to try them out, and then decide if you want the pro. I only opt for the Pro version of Advanced System care.
